Background: CC-4560 (lf5-2 mt+)
Origin: Made by transforming CC-4560 cells with exogenous wild-type LF5 gene tagged with sfGFP right in front of the stop codon.
Culture maintenance: Can be maintained on Tris-acetate-phosphate (Gorman and Levine, 1965) agar plates or slants, or grown in liquid modified Medium I (Sager and Granick, 1953; Witman, 1986).
Comment: An exogenous copy of the wild-type LF5 gene tagged with sfGFP at the C-terminus right in front of the stop codon is incorporated into the genome of this strain. Thus, the long-flagella phenotype caused by the lf5-2 allele is rescued by the wild-type transgene. CC-4560 is reported to be mt+ but our analysis indicates that it is mt-.
