This is a knock-out generated by CRISPR/Cas9 (following Greiner et al. 2017) targeting exon 19 of Cre04.g220200, inserting a stop-codon–containing sequenced. Co-transformation of pBC1 (pJR38, Neupert et al., 2009) conferring paromomycin resistance.
Knockout of CrKEA1 (Cre04.g220200) is a predicted chloroplast envelope K⁺/H⁺ antiporter and homolog of Arabidopsis AtKEA1/AtKEA2, verified by immunoblot. Simply called kea1 (and not kea1-1) in Wunder et al. 2026 (bioRxiv). The exact nature of the knock-out insertion and more described in Wunder et al. 2026.
