Phenotype: impaired motility; cannot use glutamine as N source

From a cross of CC-1128 pf27 (no longer in the collection) to a gln1 strain obtained from Bob Hodson.

This strain derives from the cross that placed gln1 on linkage group XII/XIII. Subsequent crosses established that it was on the right arm of this linkage group.

The gln mutant can be scored by inability to grow on glutamine as sole nitrogen source. The test agar can be prepared using standard recipies, omitting NH4Cl and adding 0.292 g glutamine in 5 ml sterile water per liter, after autoclaving. Best results will be obtained if highly purified agar is used, e.g. Difco Noble agar.
