Vector: pBluescript KS (+)
Origin: A HindIII/SpeI fragment extending from Cre01: 1029777 – 1024634 in Phytozome v5.6 genomic DNA sequence was cloned into pBluescript KS (+) to create plasmid pCEHC1-5.1H/S. The plasmid was modified by inserting three copies of a sequence encoding the HA epitope into the SacI site located immediately downstream of the ATG start codon. In addition, a 322 bp HindIII fragment was cloned into the HindIII site to extend the promoter region.
Insert: Wild-type copy of Cre01.g005900 (CECH1 gene), nuclear DNA, 5559bp HindIII(mutated)-SpeI fragment, 3xHA tagged at 5’ end of gene.
Comment: This plasmid can rescue the mutant phenotype of the cehc1 mutation in strain CC-4509.
