Phenotype: antibiotic resistant (erythromycin)

The ery2e mutant was isolated in the Boynton/Gillham laboratory as ery12, induced by exposure to 50 micrograms/ml nitrosoguanidine, selected on 100 microgram/ml ery-HS plates.

The ery2 mutants as a group are resistant to 100 micrograms/ml erythromycin on minimal medium, and are cold-sensitive. Please see CC-1711 for more information on the ERY2 locus.

Phenotype: impaired motility

Cells of the pf10 mutant swim with a jiggling motion and travel in small irregular circles. This abnormal swimming results from a change to a nearly symmetric flagellar beat pattern, which is conditionally dependent on light intensity. The mutant is very easy to score. A number of suppressor mutations have been characterized by Dutcher and colleagues.

The ery1b mutation was isolated in Bogorad’s laboratory in the 137c background, induced by mutagenesis with ethyl methanesulfonate. This strain has been used in many crosses by the Chlamydomonas Center and has been well-behaved, easy to score.

The ery1 mutations were shown by Davidson et al. (1974) to alter a protein of the large subunit of the chloroplast ribosome, subsequently identified by Bowers et al. as the product of the RPL4 gene. The ery1b allele results in an L4 protein that is about 30% smaller than the comparable wild-type protein.
